This project investigates the role of DNA replication origins in defining cell identity during development in eukaryotes. By studying mouse embryonic stem cells, it aims to link replication origins to gene expression and identify proteins involved in their formation.
DNA replication origins remain poorly defined in metazoans, in contrast to bacteria or S. cerevisiae.
We believe that, in eukaryotes, a differential encoding of chromosomes by DNA replication origins is instrumental for the acquisition of cell identity during development.
We wish to decipher this encoding, to determine whether it is linked to gene expression, and identify the mechanisms used to build a replication origin.
