This project investigates the initiation of RNA polymerase II transcription by creating a synthetic DNA template that mimics the assembly of the preinitiation complex. It focuses on understanding the interactions between transcription factors and modified nucleosomes, employing advanced structural analysis techniques.
Initiation of RNA polymerase II (Pol II) transcription is activated by the assembly of preinitiation complex (PIC) onto gene promoters. PIC contains the largest general transcription factor (GTF), TFIID that comprises TATA box-binding protein (TBP).
